Three Friends and I just singed up for our first endurance karting event. It's a 6 hour at Atlanta Motorsports Park in their rental karts.

I've done a couple 1.5 hour races with teams of 3. I doubt anyone will want to do longer than a 20 minute stint as you just get worn out.
Figure out a pit sign and hand signals - communication goes a long way.
Drive changes - old driver rolls out to the left and new driver hops in from the right. Far smoother.
Wear grippy gloves to help with waning hand strength as the race goes on.

I did the 24-hour kart race with Endurance Karting. Four people on my team and we won our class by 7 laps. 40-50 minute stints if I remember correctly. It was some of the most fun racing I've ever had. Plus there were some IMSA drivers in the field (not in the same class as us), so it was cool to share the track with them.
You want a good rib protector. And I totally second what Dave said about grippy gloves.
How old are you and your teammates? I wouldn't hesitate to push 1hr stints on people in their early 20s. Once they get into their 30s the likelyhood of them being whiny/old/broken is quite a bit higher.
Drink lots of water and eat plenty of healthy snacks. (fruit , energy bars) Running a kart for an extended period of time is extremely tiring. Also, be sure to remind yourself to relax and breathe, and have a Hans, or at least a neck collar on. Karts generate plenty of G's, so your neck will get a workout. (actually your entire body is going to get a workout) I run in a charity 4-hour enduro every year in Chicagoland, and it's a fantastic time.
Quality gloves and a comfortable helmet are also key. Be sure to Rain-X / De-Fog your visor so you can see clearly too.
Also remember....no one wins a 24 hour race on the first lap. You'll have idiots out there that will try....... but keep the Red Mist in check, and race smart.
Plan on a bunch of couch time in the days following the race....or a hot tub. Your body will need it!

They have a 6 week 12 race series for sprint karts at AMP. Season begins March 15. You can rent a kart for $89 per week or $475 for entire 6 week season.

If you're in the Atlanta area K1 speed also does a monthly league race which I have been partaking in, and really enjoying. The bad thing is the next K1 league race is the day after the 6 hour, and I'm thinking I may be all karted out by then.
