Ok so I am about to start building a new garage. It is going to be a 24x40 and I am going to add an interior wall at the back 10ft to have a gym and office in the garage. See photo below.

 

Is the best way to do this going to be be two mini-splits?

Will just having no door be enough to keep the office cool? Or should add I add pass through vents in the wall or something?

You're in Alabama?  A/C requirements there are going to be pretty rigorous.

You really should do an HVAC load calculation to determine what is needed.  There are a number of online calculators, you just plug in the needed values (building dimensions, amount of insulation, local temperatures, etc.) and it spits out a number.  They aren't necessarily all created equal so it's a good idea to run the numbers through several of them to make sure you get a consensus.

Dual or even tri split ductless could run off of a single outdoor unit.
