I'll be spending 27hrs in Denver next Monday into Tuesday. What's a must see while there?

Depends on your interests, but driving up to Buffalo Bill Cody’s museum will give you a great view over the city & possibly some elk sightings along the way.
State capital - nice downtown area. We used to go a lot for work. Once we headed west a bit and rode the Alpine Slides. Pikes Peak might be too far?
Where u staying? Buy weed?

Either with a friend or at hotel close to the Airport. No weed for me, military.
Can't really go wrong downtown.
But red rocks ampitheater, you won't see something like that anywhere else.
So they do have a train now a days to downtown.

There are a bunch of good places to eat downtown. Some good tap rooms as well if you like the craft beer scene.
When are you heading up there?
Really depends on if you're more a city person or country person. I've visited the Denver area several times, but never went into the city as I was/am much more interested in the beautiful scenery outside of town. As noted, the Red Rocks Amphitheater is spectacular to see. Some of the small state parks are really amazing too, such as Roxoborough. I went there several times and the views are stunning.
Take a drive to Golden, which isn't all that far away. If you've got a fun car, take Rt. 6 West out of Golden toward Idaho Springs. It's a car guys dream road and the scenery is incredible. Then drive up Mt. Evans.
