I don't have a favorite necessarily, depends on my mood. Yesterday it was Dumb and Dumber.
Canoe or not, I constantly fall for immature comedy.
That said, the movie that has literally made me laugh out loud the most of any movie in history has to be Horrible Bosses. I was cracking up!
Shaun of the Dead and Hot Fuzz are on my short list. Simon Pegg and Nick Frost have some SERIOUS deadpan humor skills.

+1, Run Fat Boy Run also...
Anchorman
Zombieland
Happy Gilmore
...
Ooooh, I forgot about Run Fatboy, Run. Need to watch Paul too- even though Seth Rogan is NOT at the top of my list. Also, Zombieland is amazing. Too bad Jesse Eisenberg is always such a weenie. Woody Harrelson's character MADE that movie.
In reply to mndsm:
You haven't seen Paul yet? Very funny...
Airplane
Blues Brothers
Fish called Wanda (overall it's "meh" but has some great scenes and lines)
Dumb and Dumber
Older: Animal House, Newer: The Hangover
Both define each generation's style of humor. Perfectly. Animal House is still hilarious today, and The Hangover will still be hilarious in 40 years.
Stripes, Animal House, Planes Trains and Automobiles, Fletch, Blues Brothers.
...notice the SNL common thread here?
you all are pretty much naming all mine haha
all time, have to narrow it down to Dumb and Dumber and for new? Horrible Bosses hands down!
Don't have a particular favorite either.
But I think my favorite catch-me-off-guard comedy would be 300.
Dumb comedy: Rat Race
Slapstick-ish comedy: Liar Liar
Smart comedy: Johnny Dangerously
Dramedy: Fifth Element
Guilty pleasure comedy: Legally Blonde
Life of Brian (I think it is funnier then Holy Grail)
Raising Arizona (for wackyness)
Up in Smoke (there's something about drug humor I like)
Get Him to the Greek (current drug humor)
Galaxy Quest (Spot on Trek humor)
Slap Shot and Major League
Kung Fu Panda
Best in Show
The Hangover
Blazing Saddles
Young Frankenstein

Some people my age don't even know about this movie. It's just plain good ol' silliness
Paul was epic.
Also enjoy Hot Fuzz, Shaun of the Dead, Zombieland, Galaxy Quest, Hangover, and of course the all-time greatest ever comedy:
SPACEBALLS!!!!
